HiPEG™
Many low molecular weight proteins are produced with a polyhistidine-tag for affinity purification post expression in Escherichia coli and other expression systems.
PolyTherics has developed a way of covalently conjugating one or more PEG molecules specifically to a histidine-tag; this technology is called HiPEG™. The polyhistidine-tag can still be used for purification of the protein following PEGylation. HiPEG™ is ideal for targeted PEGylation of peptides and proteins lacking accessible disulfide bonds which are thus unsuited to TheraPEG™. Other his-tagged proteins are currently in clinical development.
HiPEG™ has been successfully applied to PEGylation of a wide range of proteins, and peptides, as shown below.
PolyTherics has shown in an in vitro study that full activity was retained when a single 20 kDa PEG was conjugated to an anti-TNFa domain antibody using HiPEG™.
HiPEG™ has also been used to produce a novel version of IFN α-2a which PolyTherics is developing in-house as a biobetter product. This is the Company’s most advanced programme.
